The UB Distinguished Speakers 2022-23 series will feature actor Ken Jeong, former U.S. Ambassador to Ukraine Marie Yovanovitch, civil rights lawyer Sherrilyn Ifill and actress Selma Blair.

The University at Buffalo Distinguished Speakers Series returns for 2022-23 with a range of engaging, inspiring and entertaining speakers.

The season will feature actors Ken Jeong and Selma Blair, former US Ambassador to Ukraine Marie Yovanovitch and civil rights lawyer Sherrilyn Ifill.

All lectures will be held at 7:00 p.m. at the Arts Center Mainstage Theater, North Campus.

Lupus is a chronic autoimmune disease that attacks healthy tissue. It can cause bouts of tiredness, joint pain or swelling, skin rashes, headaches, sensitivity to light, and chest pain when breathing deeply. In extreme cases, it can be fatal when it causes damage to internal organs.

No cure exists for lupus. Doctors are deploying an array of therapies aimed at easing its symptoms and reducing the damage it can cause.

At the 2022 Fall Meeting of the American Chemical Society on August 21, 2022, Bristol-Meyers-Squibb (BMS) presented the results of promising research regarding a pill that BMS hopes can directly treat systemic lupus erythematosus ( THE). SLE is the most common form of lupus.

The new pill is called afimetoran, and it reduced lupus-like symptoms, reversed organ damage and prevented deaths in mice.

BMS has begun phase 2 human clinical trials of afimetoran. Phase 2 trials test the effectiveness and side effects of a drug.

Albert Roy is currently Executive Director of Lupus Therapeutics of the Lupus Research Alliance and will become President and CEO of the Alliance in September 2022. He described the effects of SLE on Medical News Today:

“Up to 90% of people with lupus will have inflammation or swelling of the joint lining, causing stiffness and pain, most commonly in the hands and wrists. Most people with SLE have unexplained fevers .

Fatigue from lupus, Roy noted, can be “severe enough to have a significant impact on patients’ quality of life, including reducing their ability to function at home or at work.”

“About half of people with lupus have a red ‘malar’ rash that can appear on the cheeks and butterfly-shaped bridge of the nose or other parts of the body, and can be painful or itchy. Because many people with lupus are photosensitive, rashes often first develop or worsen after exposure to sunlight.

Lupus can also lead to cardiovascular disease, kidney disease, and stroke.

“Specifically,” Roy said, “people with lupus are at increased risk for atherosclerosis — the deposition of fats and cholesterol [plaque] along the wall of the arterial wall. In some people, inflammation can occur in the heart itself (myocarditis and endocarditis) or in the membrane around it. Endocarditis can damage heart valves, which can lead to heart murmurs.

“When the disease affects the kidneys, it is called lupus nephritis, and patients usually need intensive drug treatment to prevent permanent damage. Lupus can also attack the brain or central nervous system, which can cause seizures or a stroke,” he continued.

To add to the challenge of treating lupus symptoms, Roy said, “there is no single test to definitively diagnose lupus, and it could take months or even years to be sure.”

Diagnose lupus

“Typically, your doctor will perform a complete history and physical examination, including blood tests. The doctor may also perform skin and kidney biopsies (removal of tissue samples which are then examined under a microscope) to establish a diagnosis.

Other common types of lupus, Roy said, are:

  • Cutaneous lupus, which causes a rash or lesion on the skin, usually when exposed to sunlight
  • Drug-induced lupus, similar to SLE, which is caused by an overreaction to certain medications. Symptoms usually go away once the drug is stopped.
  • Neonatal lupus, which occurs when an infant acquires autoantibodies from their mother with SLE.

Afimetoran inhibits the function of the cellular proteins TLR7 and TLR8.

TLR7 and TLR8 trigger the immune system, usually in response to foreign RNA. However, in people with lupus, they activate the immune system when they mistake the individual’s own RNA for a threat, causing a range of lupus symptoms.

“It is well documented,” says Roy, “that aberrant activation of TLR7/8 is potentially pathogenic and linked to the progression of certain autoimmune diseases such as lupus. Previous efforts to develop TLR7/8 inhibitors have been largely unsuccessful due to the challenge of producing a small molecule inhibitor for these difficult targets.

Dr. Alaric Dyckman, director of Bristol Myers Squibb, recalled an important finding from their research.

“The first remarkable discovery was the identification of small molecules that could bind directly to TLR7 and TLR8 and inhibit their function, confirming that they are valid drug targets for this modality,” he said. MNT.

BMS identified the molecules by screening the company’s huge collection of compounds for anything that might block TLRs 7 and 8.

Dr. Dyckman described the BMS collection:

“The Compound Collection is a physical inventory of a large number of purified samples that have been preserved for many years and are carefully stored for continued use. The samples in the collection come from materials prepared in-house, as well as from samples purchased from external sources For the TLR screening, 1.25 million samples from this collection were tested for their inhibitory capacity in cell-based assays.

Inhibition of TLRs 7 and 8 should pose no new problems, Dr. Dyckman believes:

“TLR7 and TLR8 function as part of the innate immune system, providing surveillance against pathogens. They are not alone in this role, and a range of other proteins in the body perform similar and overlapping protective functions. This redundancy supports the idea that a TLR7 and TLR8 inhibitor could be administered without undue concern for broad immunosuppression.

“For mouse models of lupus, we were able to show that not only could we prevent the development of lupus-like symptoms in animals receiving our compound treatment before disease onset, [but] we could [also] actually reversing symptoms in animals that had established disease, preventing the lethality associated with these very challenging models,” Dr. Dyckman said.

“Such a reversal had not been demonstrated with other mechanisms that we had assessed in animal models of the disease prior to this work,” he added.

The researchers also found that afimetoran worked well with corticosteroid treatments in in vitro studies and in mice. This is important because doctors often prescribe such steroids to treat inflammatory symptoms of lupus.

“In Phase 1 human clinical trials,” said Dr. Dyckman, “we were very pleased to find that across a range of single and repeat doses, afimetoran was well tolerated.”

With a single 24-hour dose, he said, “afimetoran, once daily, was able to almost completely block signaling via TLR7 or TLR8 with very low doses.”

BMS will continue to monitor safety during phase 2 trials.

“The Lupus Research Alliance and Lupus Therapeutics,” Roy said, “are proud to partner with BMS, Amgen, Lilly, UCB, Biogen and Vera Therapeutics to advance and evaluate their novel therapeutic approaches.”

When it comes to staying healthy, medical experts always talk about diet and exercise. They don’t talk about sleep, but in my experience sleep has been among the best medicines. When I sleep well, I get sick less often, think better and have more energy. Studies confirm that insomnia (difficulty sleeping) is a big problem for people with multiple sclerosis (MS) and is closely linked to one of the worst symptoms, fatigue.

I have often written about sleep for diabetes publications because poor sleep worsens blood sugar control. I find that a night of poor sleep leaves me a bit depressed and anxious, which studies show is a common pattern in people with MS.

They say that MS is caused by our immune system and that sleep is vital for good immune function. Sleep is when the immune system can get to work without all the demands of waking life. People with insomnia have higher levels of inflammatory immune system proteins such as tumor necrosis factor, and inflammation is an important part of MS. So most of us can benefit from more sleep.

The problem is that MS can interfere with sleep, because we tend not to move as much while awake as healthy people. Thus, our body does not get tired, even if our brain does. And if your body isn’t tired, it won’t sleep (unless you’re on drugs). Other MS symptoms, such as pain, muscle spasms and bladder problems, can also interfere with sleep.

When we go to bed and can’t sleep, or wake up for some reason, just lying there thinking or tossing and turning won’t help. If you have good ideas that are more important than sleep, write them down so you don’t forget them! (It happens to me sometimes.)

But if you really want to fall asleep, don’t give it more than 15 or 20 minutes. If you’re still awake, get up and do something useful, or stay in bed and do something really boring. Nothing puts me to sleep faster than reading a boring book or doing a silly puzzle, but doing the dishes or doing gentle exercise seems to lead to better rest.

Vigorous exercise before bed can keep some people awake, but vigorous exercise isn’t really a thing for many people with MS. Gentle exercises – I do seated qi gong or very light (2lb) arm exercises – really help me relax and fall asleep. Sitting at a computer scrolling through social media makes it harder.

Sleep experts talk about having a bedtime ritual, which means regular behavior that tells your body “time to relax.” Often this includes turning down the lights, remembering good things that happened that day, making a list of what you will do tomorrow, some gentle stretching, maybe drinking herbal tea, warm milk, listening to music. peaceful music or relaxation sounds, or eat a snack. (No caffeine!)

I find it helpful to remember my loved ones and give thanks for what I have received. If possible, resolve small issues, such as a misunderstanding with a spouse that you might be worried about. As marriage counselors say, don’t go to bed angry.

It is important to have a quiet, dark and comfortable place to sleep. Unfortunately, not everyone has this, but if you can create such space, you’ll definitely sleep better and feel better.

It helps to have a sleep schedule. I used to work night shifts as a nurse and sometimes wonder if that contributed to my MS. Some studies have shown that shift work can affect many people’s immune systems. Now I try to fall asleep, go to bed at the same time, turn off the light after 30 or 60 minutes of reading. I try to go outside every day, especially on sunny days, because the daytime sun helps people sleep.

This schedule has worked very well, except that I have to get up to urinate (actually autocath myself) once a night. Then I go back to bed, but if sleep doesn’t come quickly (within 5 minutes) I get up and stretch, write or do the dishes.

And the naps? Nap time is a ritual in many cultures, and for people with MS, napping can be unavoidable. Short naps (called power naps) are great, and you might even be able to take them during a lunch break at work. I also like longer naps, but some scientists say long naps can interfere with nighttime sleep. I haven’t noticed this problem, but maybe you can see how they work for you.

I advise not to use sleeping pills. They’re not widely prescribed anymore, because they can leave people as tired as if they hadn’t slept, and they’re not safe. Especially for older people with MS, they cause too much risk of falling when we get up while still under their influence. This applies to cannabis as well as prescription drugs.

At this point in my life, sleep has become one of my favorite things. No stress, no pain. I imagine all my little cells going around repairing the damage and preparing my body for the next day. And I look forward to dreams, like trips to another world or a big movie. Sometimes in dreams I have my full abilities back, sometimes not, but they are always interesting and often teach important lessons.

We live in an anti-sleep society, where we are expected to be productive all the time. But don’t believe this hype. Sleep is great medicine.

After months of waiting, the first MCU movie of 2022 is almost here – and it’s sure to be epic. Doctor Strange in the Multiverse of Madnesscoming on the heels of Spider-Man: No Coming Home, has to be one of Marvel Studios’ most ambitious offerings yet. Just look at the slew of major characters featured in this new sequel poster.

With only 10 days – yes, really – until it hits theaters, Marvel shared one last poster for Doctor Strange 2 which spotlights Doctor Strange, his friends and foes, not to mention their many variants that will feature in the film. While most of them are decipherable, some numbers are deliberately hidden. So, fan sleuths, do your thing.

This latest poster highlights how much more of an ensemble piece this one will be than the first. strange doctor, with star appeal top billing crediting Benedict Cumberbatch (Stephen Strange), Elizabeth Olsen (Scarlet Witch), Chiwetel Ejiofor (Karl Mordo), Benedict Wong (Wong), Xochitl Gomez (America Chavez), Michael Stuhlbarg ( Nicodemus West) and Rachel McAdams (Christine Palmer).

While the others appeared prominently in the promo, Stuhlbarg’s credit here is a bit of a surprise as it suggests he’ll have a larger role in the movie than expected. It seems likely that West, a former neurosurgeon rival to Strange, could be Christine’s husband, as trailers have seen her walking down the aisle in white.

And, remember, these guys are just the tip of the iceberg as we’re expecting a slew of familiar face cameos from across the Marvel movie multiverse. So far Patrick Stewart’s Professor X and the Illuminati have been confirmed, but bets are good there will be plenty more where they came from.

Superhero movies have kept the box office afloat this year, prompting theater operators to believe that audiences may finally be ready to return to cinemas in droves.

Domestic ticket sales for the first four months of the year are still down around 40% from pre-pandemic 2019 levels, but theaters are seeing significant gains from a year ago.

Hit titles like Warner Bros.’ Paramount’s ‘The Batman,’ ‘Sonic 2,’ and Marvel-Sony’s ‘Spider-Man: No Way Home’ drove ticket sales up 365% from 2021, reaching $1.85 billion, according to Comscore data.

Theater owners tell CNBC they hope ticket sales continue to rise in the coming weeks, especially after the release of Marvel’s season-opening ‘Doctor Strange in the Multiverse of Madness’ summer blockbusters.

The past two years have suffered from lackluster summer movie slates, as lockdowns shuttered theaters and coronavirus strains kept many would-be moviegoers at home.

Studios had been reluctant to release films, fearing their new films would not turn a profit and many opted to postpone titles until 2022. Now, with many health and safety mandates repealed and consumers seemingly more comfortable returning to theaters, studios stuck to their release dates.

Theaters will see a steady stream of highly anticipated films following the release of “Doctor Strange” on May 6. ‘Top Gun: Maverick’, ‘Jurassic World: Dominion’, ‘Lightyear’, ‘Minions: The Rise of Gru’ and ‘Thor: Love and Thunder’ will hit the big screen in quick succession over a nine week period.

“It’s only fitting that a Marvel movie kicks off the 2022 summer movie season and kicks off what seems like the first ‘normal’ May Labor Day streak we’ve seen. for almost three years,” Paul said. Dergarabedian, senior media analyst at Comscore.
